Enhanced forensic DNA recovery with appropriate swabs and optimized swabbing technique

چکیده
Efficient sampling with swabs is crucial for optimal forensic DNA analysis. The recovery determined by the skill of practitioner and compatibility between applied swab surface. Here we investigate impact swabbing technique type on yield. Thirteen different from four categories (cotton, flocked nylon, small foam large foam) provided equal yields smooth/non-absorbing surfaces. Large gave higher an absorbing wood Factorial design experiments ANOVA was to study techniques cotton swabs. Two key factors efficient were found be 1) holding approximate 60° angle against surface 2) rotate during sampling. For wood, it beneficial wet heavily. results factorial used develop protocols When ten experienced practitioners sampled according these protocols, yield increased ridged plastic (around 1.25 times more DNA) (2.2–6.2 DNA). window glass, representing a surface, protocol equivalent applying individual techniques. lowered person-to-person variation plastic. In conclusion, have developed instructive three types surfaces: smooth/non-absorbing, ridged/non-absorbing smooth/absorbing. We believe that such will streamline simplify training new improve efficiency invisible residues in casework.

A Novel Improved Bioburden Recovery Method Using Swabbing Technique
The validation of surface-recovery methods is a pre-requisite for residual determination of cleaning effectiveness in process validation studies. These methods should be challenged in the laboratory using pilot-scale controlled conditions in order to evaluate the suitability for their intended use. Different swabbing techniques have been studied and compared with rinse method for 7 microorganis...
متن کاملEvaluation of Methods to Improve the Extraction and Recovery of DNA from Cotton Swabs for Forensic Analysis
Samples for forensic DNA analysis are often collected from a wide variety of objects using cotton or nylon tipped swabs. Testing has shown that significant quantities of DNA are retained on the swab, however, and subsequently lost. When processing evidentiary samples, the recovery of the maximum amount of available DNA is critical, potentially dictating whether a usable profile can be derived f...
متن کاملEnhanced Genetic Analysis of Single Human Bioparticles Recovered by Simplified Micromanipulation from Forensic ‘Touch DNA’ Evidence
DNA profiles can be obtained from 'touch DNA' evidence, which comprises microscopic traces of human biological material. Current methods for the recovery of trace DNA employ cotton swabs or adhesive tape to sample an area of interest. However, such a 'blind-swabbing' approach will co-sample cellular material from the different individuals, even if the individuals' cells are located in geographi...
متن کاملSwabbing: touchscreen-based input technique for people with hand tremor.
Interacting with touchscreen computer systems is a challenging task for elderly tremor patients, because of a lower input accuracy caused by their tremor symptoms. Previous observations by Mertens et al. suggested that continuous movements are able to lower these symptoms. In this paper Swabbing, a input method for touchscreen systems based on such motions, is presented. A user study will show ...
متن کاملThe influence of swabbing solutions on DNA recovery from touch samples.
There has been minimal research into how to best obtain DNA from touch samples. Many forensic laboratories simply moisten a swab with water and use it for collecting cells/DNA from evidentiary samples. However, this and other methods have not been objectively studied in order to maximize DNA yields. In this study, fingerprints were collected using swabs moistened with water or laboratory or com...
